Post Doc position
Genome engineering in the yeast Pichia pastoris
in Vienna / Austria for 40 hours a week
That’s the project
The yeast Pichia pastoris is an important host for the production of industrial enzymes and biopharma-ceutical proteins. Typically, the expression cassettes are integrated into the genome by homologous recombination into a predefined locus. Multicopy strains containing multiple copies of the expression cassettes are usually associated with higher productivity but have been shown to be instable sometimes.
To accelerate strain development, highly efficient methods for gene integration are needed. In this project, we aim to establish novel methods for gene integration in P. pastoris based on DNA modifying enzymes. This also includes the development and evaluation of novel means of transformation. The resulting strains will be characterized for growth and productivity in high throughput screening and bioreactor cultivations. Genome integration events will be analyzed by genome sequencing.
